Surface receptor Toso controls B cell-mediated regulation of T cell immunity.

چکیده
The immune system is tightly controlled by regulatory processes that allow for the elimination of invading pathogens, while limiting immunopathological damage to the host. In the present study, we found that conditional deletion of the cell surface receptor Toso on B cells unexpectedly resulted in impaired proinflammatory T cell responses, which led to impaired immune protection in an acute viral infection model and was associated with reduced immunopathological tissue damage in a chronic inflammatory context. Toso exhibited its B cell-inherent immunoregulatory function by negatively controlling the pool of IL-10-competent B1 and B2 B cells, which were characterized by a high degree of self-reactivity and were shown to mediate immunosuppressive activity on inflammatory T cell responses in vivo. Our results indicate that Toso is involved in the differentiation/maintenance of regulatory B cells by fine-tuning B cell receptor activation thresholds. Furthermore, we showed that during influenza A-induced pulmonary inflammation, the application of Toso-specific antibodies selectively induced IL-10-competent B cells at the site of inflammation and resulted in decreased proinflammatory cytokine production by lung T cells. These findings suggest that Toso may serve as a novel therapeutic target to dampen pathogenic T cell responses via the modulation of IL-10-competent regulatory B cells.

Recent Advances in T Cell Signaling in Aging
The immune system of mammalian organisms undergoes alterations that may account for an increased susceptibility to certain infections, autoimmune diseases, or malignancies. Well characterized are age related defect in T cell functions and cell mediated immunity. Although it is well established that the functional properties of T cells decrease with age, its biochemical and molecular nature is...
متن کاملEVALUATION OF CELL-MEDIATED IMMUNITY IN MUSTARD GAS INJURIES
Yamakido, et al. in 1986 have studied immunological parameters in poison gas workers, and a depression has been observed in immunologic response. Also, the frequency of cancer has reached as high as five-fold that of the general population. In the present study, cell-mediated immunity (CMI) was measured in three groups of Iranian mustard gas-injured patients. The first group were those wh...
متن کاملToso, a functional IgM receptor, is regulated by IL-2 in T and NK cells.
We find that the cell surface receptor Toso is dramatically downregulated by in vitro stimulation of human T and NK cells with IL-2 in a STAT5-dependent manner. The fact that IL-2 is known to prime NK and T cells for Fas/TNF-mediated activation-induced cell death (AICD) fits nicely with the original and recent descriptions of Toso as an inhibitor of Fas/TNF-induced apoptosis. In support of this...
متن کاملToll Like Receptor-4 896A/G Gene Variation, a Risk Factor for Migraine Headaches
Background: The pathogenesis of migraine involves immune-mediated mechanisms in the vascular endothelium. Toll like receptor 4 (TLR-4) is a signaling receptor of innate immunity which plays a role in various neuropathologies related to neuron inflammation. Objective: This case/control study is aimed to investigate whether TLR- 4 896A/G variation is related to migraine headaches in an Iranian po...
متن کاملToso controls encephalitogenic immune responses by dendritic cells and regulatory T cells.
The ability to mount a strong immune response against pathogens is crucial for mammalian survival. However, excessive and uncontrolled immune reactions can lead to autoimmunity. Unraveling how the reactive versus tolerogenic state is controlled might point toward novel therapeutic strategies to treat autoimmune diseases. The surface receptor Toso/Faim3 has been linked to apoptosis, IgM binding,...
